Tissue engineering (TE) and xenotransplantation
Offer Description The meniscus, a fibrocartilaginous tissue, stabilizes the knee and distributes load but has limited healing capacity, making it prone to injury. Due to the low success of meniscal repair and meniscectomy, preserving its mechanical properties is crucial. Tissue engineering (TE) and xenotransplantation, particularly using pig models, offer promising solutions. This project aims to advance meniscus TE and xenotransplantation by analyzing the genetic makeup of pig menisci. Using single-cell and long/short-read sequencing, we will build a detailed reference transcriptome, compare it with human data, and explore zonal composition, healing potential, and immune responses. In-depth analysis will be performed with the state of the art sequencing techniques – single cell approach combined with short and long-read sequencing. Merging this two types of data will allow for building a detailed pig menisci reference transcriptome, which will include whole polyA transcripts with unique splicing variants. Obtained data will be compared with publicly available human data in terms of divergent zonal composition, cell interactions, as well as chondrocyte and epithelial cell types and functions. On the other hand spatial transcriptomics will allow for unrevealing internal organization of cells and define spatial location of those involved in expressing epitopes potentially triggering immune system of the xenograft recipient. Offer Description Vice-Rector for Finance and Research Projects School of Natural Sciences at the Adam Mickiewicz University, Poznań announces a competition for the position of: assistant professor (post-doctoral researcher) at the Faculty of Biology in the project OPUS “Multiomics and functional analyses in type 2 diabetes: targeting altered pathways and metabolic rearrangements.” (number UMO-2024/53/B/NZ4/03103). We are looking for a motivated researcher for the position of a postdoctoral fellow at the Laboratory of Stem Cell Biology and Regenerative Medicine, Adam Mickiewicz University in Poznan, in an OPUS project funded by the National Science Center, NCN. The successful candidate will study changes in pancreatic organoids derived from patients with type 2 diabetes using multi-omics techniques. The research will be conducted using state-of-the-art techniques, including the differentiation of human stem cells into pancreatic beta cells and the production of organoids, advanced genome editing during differentiation, single-cell RNA sequencing, proteomics and lipidomics, functional assays, and microscopic imaging. Mastering all of these techniques is not necessary, as the project will be carried out in collaboration; however, there is an opportunity to learn more about them. The knowledge gained will be crucial for the development of cell and gene therapies for people with diabetes. Michał Gdula to work on leveraging epigenetics to improve the effectiveness of genetic therapies for diseases caused by nucleotide repeat expansions (including myotonic dystrophies), within the framework of the International Research Agenda (MAB) FENG grant funded by the Foundation for Polish Science (FNP). Work should aim at writing and defending a doctoral dissertation. Dr. hab. Michał Gdula’s group is one of four teams participating in the TREAT-EXP project, led by Prof. Krzysztof Sobczak and funded by FNP with PLN 30 million over four years. The overarching goal of TREAT-EXP is to build a research platform to develop therapeutic strategies and identify active compounds targeting pathogenic products of mutated genes—either toxic RNA or toxic proteins—responsible for DM, FXPAC, and ALS. The goal of Michał Gdula’s team is to increase the efficacy of therapies for DM1 and DM2 by modulating epigenetic regulation using modified antisense oligonucleotides (ASOs). DM1 is caused by a CTG expansion in the 3′UTR of the DMPK gene, while DM2 results from a CCTG expansion in an intron of the CNBP gene; the resulting toxic repeat-expanded RNA forms nuclear foci and sequesters MBNL proteins, leading to widespread alternative splicing defects and multiple multisystem symptoms. The Researcher assistant’s tasks will include characterizing the epigenetic landscape of DM-associated genes and mapping their regulatory elements in cellular models using genomic methods (e.g., ATAC-seq, CUT&RUN, Capture-C, Micro-C). Next, in collaboration with—and drawing on the expertise of—Prof. Sobczak’s group, ASOs will be designed to target selected regulatory elements in order to reduce DMPK/CNBP expression or increase the expression of MBNL genes. Our research explores how inflammatory and metabolic adaptations shape TAM function during tumor progression and how targeting mitochondrial function and reactive oxygen species (ROS) signaling in tumor and immune cells can enhance anti-tumor responses. Particular emphasis is placed on cellular adaptation to physiologically relevant oxygen conditions (physioxia vs. hypoxia). The position is funded by Horizon Europe (Call: Horizon-Miss-2024-Soil-01) and part of the project “ProPollSoil” (Understanding and managing soil health impacts to protect soil-dependent pollinators). The project focuses on soil-dependent pollinators, e.g. ground-nesting bees, and their relationship to soil conditions. In collaboration with soil scientists, it will examine the extent to which land use and climatic conditions influence soil-dependent pollinators. Samples will be collected in Germany as well as European project partner countries and include sampling for chemical and physical soil properties, soil fauna, and pollinators. New methods to sample and monitor pollinators will be developed. Tasks: Development and testing of new methods (e.g. eDNA, Camera and drone imagery, deep-learning based image analyses) for monitoring soil-dependent pollinators, especially ground-nesting wild bees Application of new methods to investigate ecological research questions within the ProPollSoil project, e.g., analyzing whether ground-nesting wild bee diversity differs across land-use categories and soil types scientific services in research, including authoring and publishing peer-reviewed scientific articles and presenting research findings in team meetings and at scientific conferences The position is limited to a time period deemed adequate for the completion of a doctoral degree.
