The Department The Department of Laboratory Medicine has a strong translational focus and housing groups with leading expertise in tools spanning epigenomics and transcriptomics, functional experimental animal models, clinical samples and state-of-the-art knowledge in genome engineering and delivery. Overall ambition to address both fundamental clinically relevant questions in autoimmune conditions, monogenic diseases, and cancers. The research group Metabolites and subcellular protein compartmentalization ultimately regulate epigenetic remodeling, transcription, and cell function. In metabolically highly active cells, this regulation can become disrupted since metabolites and proteins are simultaneously required in parallel but separate cellular compartments. The Carlström laboratory investigates the fundamental mechanisms underlying metabolic–epigenetic interplay and the role of subcellular protein compartmentalization. To do this, we use various tools, including iPSCs, primary cells, and animal models, with a specific focus on glia cells and their roles in regeneration and cancer. To unravel these intricate interactions, we employ a wide range of cutting-edge technologies for cell manipulation and high‑resolution profiling, helping to overcome the current lack of suitable methodologies in this emerging field. The research group The Rydén and Mejhert Laboratory investigates how alterations in human white adipocyte function contribute to cardiometabolic disorders, including type 2 diabetes, dyslipidemia, and atherosclerosis. Our multidisciplinary team of postdoctoral fellows, PhD and master’s students, and technical staff conducts cutting-edge translational research. We are equipped with state-of-the-art facilities, including a comprehensive preclinical laboratory at KI Campus Solna (Biomedicum) and a dedicated clinical research unit at Karolinska University Hospital, Huddinge, enabling integration from basic science to clinical application. The doctoral student project and the duties of the doctoral student We are seeking a highly motivated and ambitious PhD student to join our laboratory. The doctoral project will focus on understanding how post-transcriptional gene regulation shapes cellular identity and metabolic function in human white adipose tissue. In particular, the project aims to define how alternative polyadenylation (APA) regulates gene expression programs across adipose cell types and how these mechanisms contribute to metabolic health and disease. Using state-of-the-art single-cell and single-nucleus RNA sequencing datasets, the student will first develop computational approaches to quantify APA dynamics and infer cell-state trajectories based on polyadenylation site usage. This includes building analytical frameworks to model transcript isoform selection and to reconstruct cellular transitions driven by post-transcriptional regulation. Building on these approaches, the student will generate an integrated atlas of APA across human adipose tissue, identifying cell-type-specific APA events and their alterations in response to metabolic stress such as obesity and weight loss. Based on these findings, candidate regulators of APA will be identified and functionally tested in human adipocyte models. Using genetic perturbation approaches such as siRNA-mediated knockdown or CRISPR-based gene modulation, the student will assess how specific factors influence APA dynamics and downstream metabolic processes. Readouts will include 3′ RNA sequencing to quantify polyadenylation changes, as well as functional assays measuring adipocyte differentiation, lipid accumulation, and insulin-stimulated glucose uptake. In this way, the project integrates large-scale computational analysis of single-cell omics data with mechanistic experiments in human adipocytes to directly link RNA processing to metabolic function. The student will receive broad training in bioinformatics, single-cell data analysis, and computational pipeline development, alongside experimental approaches including human cell culture, gene perturbation techniques, and metabolic phenotyping, and will work closely with an interdisciplinary network of collaborators in systems biology and RNA biology. Do you want to contribute to improving human health?  The Nordic Brain Network (NBN, PI: Prof Miia Kivipelto) is part of the section of Clinical Geriatrics, Center for Alzheimer Research, NVS. The NBN is a multidisciplinary research team at KI, but team members are also present at KUH, Finland and UK. NBN research focus is on prevention, diagnosis and treatment of ADRD. The NBN conducts cutting edge biomedical, epidemiological, observational and qualitative research, as well as randomized controlled trials (RCTs) and implementation studies for dementia risk reduction, and is part of several international collaborations. Prof Kivipelto team collaborates with other teams at NVS and the section of Clinical Geriatrics, including Prof Eric Westman team, which leads innovative research on neuroimaging in relation to ADRD. Co-led and participated projects included large national and international initiatives, including AD-RIDDLE, ACCESS-AD, TraCeDem, World-Wide FINGERS. Prof Kivipelto has also launched the Brain Health Clinic (KUH Solna) which is part of the growing multinational network on Brain Health Services (BHS) for the Prevention of Cognitive Impairment and Dementia (ICOPAD). Overall NBN aim is to develop and promote implementation of risk reduction approaches for ADRD, based on combination of non-pharmacological and pharmacological interventions, within the framework of precision medicine. Your mission  This Assistant Professor position offers a unique opportunity to build a strong academic profile – primarily by developing your independence as a researcher, while also gaining valuable teaching and pedagogical experience. You will be expected to develop and lead a research group in the field of ADRD risk reduction, including expanding the BHS collaborative work, and contributing in coordinating and executing tasks related to ongoing projects with aligned scopes. PhD Position in Glioblastoma Immunology

The Sten Linnarsson Lab, part of the Unit of Molecular Neurobiology at the Department of Medical Biochemistry and Biophysics (MBB), invites applications for a PhD student position. Our lab has a world-leading reputation for pioneering research on brain development using cutting-edge single-cell technologies. We are now focusing on unravelling the biology of glioblastoma (GBM), the most aggressive brain tumour, to identify new treatment targets The research group Our research focuses on the cellular composition of the human brain and brain cancer. We use single-cell analysis to discover cell types, cell states and lineages in the healthy human brain. We leverage these molecular atlases to map cells from human brain cancer, hoping to discover unique cell states present in tumors that could be targeted for therapy. To achieve these goals, we have developed technologies for extremely sensitive and accurate detection of RNA and DNA in single cells. We use advanced molecular biology, large-scale DNA sequencing, microfluidics and imaging. We use AI/ML to model molecular states, and build synthetic vectors to target and manipulate cell states in vivo. Ultimately, we hope to develop effective programmable therapies to treat brain cancer. The doctoral student project and the duties of the doctoral student This PhD project pursue to investigate immune dysfunction in glioblastoma (GBM) and develop novel immunotherapeutic strategies. Despite advances in cancer immunotherapy, GBM remains largely resistant to immune-based treatments, primarily due to its highly immunosuppressive tumour microenvironment that impairs antigen presentation and limit effective T-cell responses as tumour-infiltrating T cells exhibit dysfunctional behaviour. The first aim of the doctoral studies is to characterize the infiltrated T-cells phenotypes and investigate how their functional states are influenced by the local microenvironmental niches. To study the spatial immune landscape and T-cell clonal dynamics of GBM, single-cell transcriptomics and TCR sequencing using brain-patient samples will be performed and analysed. Building on these findings, the candidate will evaluate a tumour reprogramming strategy designed to restore antigen presentation and enhance anti-tumoral T-cell responses. To this end, the student will establish a humanized orthotopic GBM mouse model using CD34⁺ hematopoietic stem cell engraftment and patient-derived tumour organoids to study tumour-immune interactions and assess possible therapeutic efficacy in vivo. The candidate will perform a broad range of experimental tasks, including isolation and processing of human and murine tissues, advanced multiparametric flow cytometry, in vitro functional assays such as co-culture and cytotoxicity assays, and in vivo work using mouse models.
